방문교수 Steffen Vanneste 세미나 강연

 

2018년 5월 14일, 겐트대 본교 방문교수 Steffen Vanneste는 식품 미생물에 대한 흥미로운 강연을 진행했다. 옥신은 식물에서 발견되는 희귀한 호르몬으로써 식물 성장에 영향을 주고, 이 호르몬의 작용 양식을 이해하기 위해서는 옥신의 이동 메커니즘을 파악하는것이 중요하다. 세포 밖으로 전송된 옥신은 원형질 막에 위치한 수송체를 안정시켜 식물 성장에 영향을 준다. 우리는 옥신이 유도한 칼슘 신호가 이 과정에 역할을 할 수 있을거라고 가정하여 옥신 유도 칼슘 신호를 다루기 위해서 신호가 생성하는 데에 억제하는 작은 분자들에 대한 화학-유전적 스크린을 통해 조사를 했고, 억제제들을 통해서 칼슘 신호가 옥신 수송체의 자동조절 역학에 많은 영향을 끼친다는 것을 발견했다. 또한 칼슘이 감지되는 과정에 대하여 조사를 하였는데 칼슘에 의존하는 키나아제들 중 옥신 수송체를 안정시키는 강한 효소들이 존재한다는 것을 발견하였으며 이것들이 옥신 이동에 영향을 주는 잠재적인 대표 성분이었다. 이 경로에 따른 더 많은 과정 묘사를 통해서 식물 성장의 내부 과정을 이해할 수 있으며, 더 나아가 장기적으로 농업 특성이 개선될 수 있는 전략을 세우는 데에 도움될 것이다.

 

Steffen Vanneste 교수는 공학박사학위를 받았고 현재 겐트대학교와 VIB 연구소 식물시스템생물학학부에서 연구원으로 연구활동을 진행중이다.

원문: Calcium- The Missing Link in Auxin Action

Auxin is an elusive plant hormone, affecting nearly every aspect of plant development. Key to understanding auxin's mode of action is unraveling the mechanisms of self-regulated auxin transport. In this model, auxin that is transported out of the cell stabilizes the auxin transporters at the plasma membrane, thus generating a self-organizing subcellular asymmetry in auxin transport capacity that drives plant development. However, the mechanism of how auxin regulates auxin transport asymmetry is not well understood. We postulated that auxin-induced calcium signaling could represent an important piece in the puzzle. to be able to manipulate auxin-induced calcium signaling, we performed a chemical genetic screen for small molecules that inhibit auxin-induced calcium signaling. Via these inhibitors, we found that calcium signaling severely impacts on the self-regulated dynamics of auxin transporters. Following up on this finding we zoomed in on the corresponding calcium-sensing machinery. Using a conditional induction system, we found that a subgroup of calcium-dependent kinases has a strong potency to stabilize auxin transporters at the plasma membrane, possibly representing a novel element in the pathway of self-regulated auxin transport. Further characterization of this pathway will lead to a better understanding of the inner workings of plant growth and development which, in the long term, will allow designing strategies to improve agricultural traits.
